The Flash apresenta a história de Barry Allen (Grant Gustin, de 90210 e Glee), assistente da polícia forense que, após sofrer um acidente no laboratório da polícia, se torna o herói Flash, o homem mais rápido do mundo.

No elenco do episódio piloto também estão Jesse L. Martin (Law & Order, Smash), Candice Patton, Tom Cavanagh (Scrubs, Royal Pains, The Following), Carlos Valdes, Danielle Panabaker (Necessary Roughness, Justified), Rick Cosnett (The Vampire Diaries), Michelle Harrison (Emily Owens M.D., Continuum), Patrick Sabongui e John Wesley Shipp, ator que interpretou Barry/Flash na série produzida na década de 1990.
